Cell Culture Workflow
As a cell culture scientist, you never stop thinking about the viability of your cells. Successful cell cultures and downstream experiments can be rapidly destroyed by problems like cell line misidentification and microbial cross contamination. Liquid Handling and Media Preparation
In this fundamental but crucial step culture media and other reagents such as phosphate buffered saline (PBS) are prepared. The laminar flow cabinet must be cleaned to ensure sterility and a suitable container for media and reagent storage is needed.
Cell Maintenance
During this step, cells are transferred into the laminar flow cabinet and are passaged into a new culture vessel for further maintenance or use in a cell assay. Centrifugation is generally used to remove and refresh expired cell culture media.
Cell Incubation
The incubation step gives cells time and the optimal environment to facilitate cell proliferation. Incubation is typically performed over multiple days or weeks at 37°C. Cells will be inside a culture vessel held within an incubator during this stage. In the case of roller bottles, roller apparatus would also be required.
Cell Cryopreservation
The aim of cryopreservation is to freeze cells in liquid nitrogen for later revival. Expired media is removed through centrifugation and the cell pellet is resuspended in a solution containing DMSO. Cells are then transferred to a cyrogenic vial which is designed to safely house cells with liquid nitrogen.
